Vapor-Phase Sterilization
Useful when sterilizing several tubes of seeds.
Use microfuge tubes marked with pencil instead of pen (pen will disappear). Poke a small hole into the top of the tube. Put seeds into tubes these tubes. Do not fill tube with seeds past the .1ml line on the tube.
Place tubes closed (but with the hole in top of tube to let in gas) in an approximately 8L bell jar (under a fume hood) containing a beaker with 100ml of household bleach (5.25% (w/v) sodium hypochlorite). Add 3.3 ml of glacial hydrochloric acid carefully to the bleach and quickly seal the jar.
Keep seeds in contact with the chlorine gas for several hours (4h-overnight).
After sterilization, keep tubes open in sterile fume hood for at least 20 minutes to disperse trace amounts of chlorine gas. Then close tubes for storage or plate seeds out for selection.
Note: Be careful. You are working with chlorine gas. Keep everything in the hood and close the bell jar top as quickly as possible upon addition of the hydrochloric acid. After sterilization it is a good idea to leave the beaker of bleach + hydrochloric acid in the fume hood overnight before dispensing in the sink.
