- Load both sides of the hemacytometer with the cell suspension - covering both counting grids.
- Count the number of cells in 5 out of 9 squares from each counting grid (count a total of 10 squares).
- Divide the total number of cells counted from 10 squares by 10 to get the average number of cells per square.
- Multiply the average count per square by 104 to get the number of cells per ml of the original cell suspension.
- Determine the grand total number of cells available for subculturing, multiply the number of cells per ml by the total number of mls of cell suspension.
