摘要:建立了手性柱HPLC法测定盐酸贝那普利的中间体(R)一a一羟基苯丁酸乙酯中的(S)一a一羟基苯丁酸乙酯。采用Chiralcel OD—H色谱柱,流动相正己烷一异丙醇(体积比90:10),流速1.0mL/min,检测波长254 nm,进样体积20 μl,按外标法以峰面积计算(R)一a一羟基苯丁酸乙酯中的(S)一a一羟基苯丁酸乙酯的含量: (S)体的线性范围5.5~23.8μg/mL,检出限0.47μg/mL(S/N=3),回收率为96%~103% ,精密度RSD为2.1%(n=6) 关键词:盐酸贝那普利;(R)-a-羟基苯丁酸乙酯;(S)-a-羟基苯丁酸乙酯;高效液相色谱;手性分离 中图分类号:0657.72;R917 文献标识码:A 文章编号:1004—4957(2005)01一0109—02 Separation and Determination of Optical Isomers of Ethyl —Hydroxyphenyl—butyrate by Chiral HPLC ZHANG Yu—ying CHEN Guo—cong (1.Guangzhou Institute for Drug Control Guangzhou 5 10160.China:2.Department of Pharmacy) ,Guangdong College of Pharmae.Guangzhou 5 10224,China) Abstract:A simple and rapid method for the determination of optical isomers Of ethyl a—hydroxyphenylbu—tyrate by chiral HPLC was developed.A Chiralcel OD—H column Was used with a mobile phase of n—hexane—isopropanol(90:10 by volume)at the detection wavelength of 254 nm。The external standard method Was employed to determine the content of ethyl(S)一a—hydroxy phenylbutyrate in ethyl(R)一a—hydroxy phenyl butyrate by peak areas。The linear range Was found to be 5.5~ 23.8μg/mL and the recovery is 96%~103%。The detection limit is 0.47μg/mL and the RSD is 2.1% (n=6)。 Key words:Benazepril;Ethyl(R)一a—hydroxy phenylbutyrate;Ethyl(S)一a—hydroxy phenylbutyrate;HPLC;Chiral separation 在很多手性化合物中,两个互为对映体的化合物在生物活性、转运机制及代谢途径等方面常常差异较大,因此,对手性化合物的对映体进行分离、分析具有重要意义。 盐酸贝那普利(洛丁新,benazepril)是一种不含巯基的强效、长效血管紧张素转化酶抑制剂,在体内水解成有活性的代谢物贝那普利拉而起作用。口服后作用迅速,可用于各型高血压和充血性心力衰竭患者。
